It centers the view on MoT, and (at least in TMO), configures your torpedoes to run at the depth of the target's keel.

So getting the torps to hit a set point is a matter of putting the cross hairs over that point and firing, by which time the target has moved.

Auto targeting will up date the firing solution to where you have the scope pointed, all locking does is track the MoT and set the depth of the torpedo that is active. All the others will stay at the depth you have set or default you haven't set them.
