But wouldn't Comic Sans mean not funny?

Sans meaning "without"

So for the joke to work, the letter needed to be read very seriously perhaps with a funny topic instead of a serious topic read funny.

Comic Sans means Comic Book like font without a serif (the little lines at the end of the letter stroke).

There is not anything inherently funny about it, especially since its designer based it on the fonts used in the (later) Batman and Watchman comics. People just tend to think of the font as something funny because it is misused so much.
