Some points from interview with Ridley Scott on Wall Street Journal:

The movie is about a higher being who challenges the gods.
The last 8 minutes of the film 'evolve into a pretty good DNA of the Alien one'. In other words, the movie ends where Alien picks up.
Sigourney Weaver doesn't make any kind of cameo appearance.
The movie was shot in 3D with CGI.

Just saw a leaked cameraphone version of the trailer. Even in low-res, you can see a couple good shots of a spaceship that looks identical to the derelict in Alien, as well as a brief glimpse of the Space Jockey rising up from beneath the floor.

If this isn't an Alien prequel, then it's surely the greatest Alien rip-off of all time!

Just watched the trailer now from NZ herald didn't know it was already in this thread, I came here to put it up. Prometheus looks damn good, can't wait to see the full movie.
