I highly recommend NYC.
It is both lightly defended, and stocked with tonnage.
If you go, expect to break the 50 000 mark easily.
I finished my NYC / East Coast with 108 000 after expending all of my deck gun, and all torpedoes.
Expect to find a Ceramic type ocean liner (14000 tons if memory serves), several Modern tankers, several liberty ships, and an assortment of tramps and medium cargos.
You will need to cross a subnet at the start of the harbour channel guarded by two bunkers.
At night with some poor weather, you can cross it easily with decks awash or a depth of about 8 meters.
I've crossed it in daylight / glass seas by lowering both periscopes into their housings, a depth set at 10 meters, and just scraped across the very top with no damage to any compartments (meaning, hull structural damage, but not serious, maybe 1-5 %).
I've also slipped through in daylight by using the external view underwater, and taking the boat fully submerged between two subnet post-dealy's, suffering no damage. Its kind of cheating though if you can slow down and use the external camera, but whatever floats your boat.
The harbour itself is patrolled by two torpedo boats and a docked destroyer.
I recommend expending torpedoes on the way in, heading up the NE channel past the harbour to surface and restock torpedoes from the external stores, and then shooting more targets on the way out.
If you are carefull you can spend your torpedoes wisely. There is no need to worry about escorts so don't be afraid to let vessels hit with single torpedoes on the first run flood and sink on their own while you go to reload. If they don't appear to by listing significantly / sunk on the exit run, put your aft tubes into them.
