Drilling through a piece of wood will blow out a chunk on the exit side.

Try it on your test board and you'll see. Same thing will happen with certain drywall materials no matter which side you start drilling on.

That's a big desk. Is it made of pressed wood and laminated veneer?

You will be needing an extra set of hands to assist in assembly. Big pieces can be quite awkward.
